Unit I

  • A brief introduction Medicinal Chemistry
  • Physicochemical properties of the biological actions
  • History & development of medicinal chemistry
  • Ionization Solubility partition coefficient, hydrogen bonding Chelation, protein binding, Optical & Geometrical isomerism
  • The principles of drug metabolism phase I & Phase II
  • Drug metabolism is affected by stereochemical aspects

Unit 2

Drugs that affect the Autonomic Nervous system.

  • Neurotransmitters Biosynthesis & breakdown of catecholamine. Adrenergic receptors & their distribution.
  • Sympathomimetic agents- Sympathomimetic agents with direct action Nor-epinephrine Epinephrine, Phenylephrine, Dopamine, Methyldopa, Clonidine, Dobutamine Isoproterenol Terbutaline, & Salbutamol. Bitolterol Naphazoline Oxymetazoline & Xylometazoline. Indirectly active agents are Hydroxyamphetamine, Pseudoephedrine, and Propylhexedrine. Agents with a mixed mechanism Ephedrine, Metaraminol.

Adrenergic Antagonists:

  • Alpha-adrenergic blockers- Tolazoline, Phentolamine, Phenoxybenzamine, Prazosin, Dihydroergotamine, Methysergide. Blockers of
  • beta-adrenergic receptors- SAR beta-blockers Propranolol, Metibranolol, Atenolol, Betaxolol, Bisoprolol, Esmolol, Metoprolol, Labetalol, Carvedilol.

Unit 3

  • Neurotransmitters of the Cholinergic family: biosynthesis & catabolism of Acetylcholine. Cholinergic receptors (Muscarinic & Nicotinic) & their distribution.
  • Parasympathomimetic agents: SAR Parasympathomimetic Agents
  • Direct-acting Agents: Acetylcholine, Carbachol, Bethanechol, Methacholine, Pilocarpine.
  • Indirect-acting inhibitors of Cholinesterase (Reversible & irreversible): Physostigmine, Neostigmine, Pyridostigmine, Edrophonium chloride, Tacrine hydrochloride, Ambenonium chloride Isofluorphate, Echothiophate iodide, Parathion, & Malathion.
  • Cholinesterase reactivator: Pralidoxime chloride.

Cholinergic Blocking Agent:

  • SAR of cholinolytic substances Alkaloids with a solanaceous structure & analogs: Atropine sulfate, Hyoscyamine sulfate Scopolamine hydrobromide Homatropine hydrobromide Ipratropium bromide. 
  • Synthetic cholinergic blocking agents: Tropicamide, Cyclopentolate hydrochloride, Clidinium bromide Dicyclomine hydrochloride Glycopyrrolate, Methantheline bromide, Propantheline bromide Benztropine mesylate Orphenadrine citrate Biperidine hydrochloride Procyclidine hydrochloride Tridihexethyl chloride Isopropamide iodide Ethopropazine hydrochloride.

Drugs that act upon Central Nervous System:

  • Sedatives & Hypnotics: Benzodiazepines, SARs of Benzodiazepines, Chlordiazepoxide, Diazepam, Oxazepam, Chlorazepate, Lorazepam, Zolpidem, Alprazolam. Barbiturates, SAR of barbiturates Phenobarbital, Barbital, Mephobarbital, & Amobarbital. Butabarbital & Pentobarbital. Secobarbital Other: Imides & imides Glutethmide. Alcohol & its carbamate derivatives Meprobomate, Ethchlorvynol. Aldehyde & its derivatives Triclofos sodium Paraldehyde.
  • Antipsychotics Phenothiazines: SAR of Phenothiazeines Promazine hydrochloride & Chlorpromazine hydrochloride, Triflupromazine, Thioridazine hydrochloride, Piperacetazine hydrochloride, Prochlorperazine maleate, Trifluoperazine hydrochloride. Ring Analogues to Phenothiazeines: Chlorprothixene, Thiothixene, Loxapine succinate, Clozapine. Fluro butyrophenones: Haloperidol, Droperidol, Risperidone. Beta amino ketones Molindone hydrochloride. Benzamides: Sulpieride.

Drugs that act on the Central Nervous System:

  • General anesthetics: Inhalation anesthetics Halothane, Methoxyflurane, Enflurane, Sevoflurane, Isoflurane, Desflurane. Barbiturates that have ultra-short-acting properties: Methohexital sodium, Thiamylal sodium, and Thiopental sodium. Dissociative anesthetics Ketamine hydrochloride. Analgesics for non-narcotic & narcotic use. Morphine & related drugs SARs of Morphine analogs, Morphine sulfate, Codeine, Meperidine hydrochloride, Anilerdine hydrochloride Diphenoxylate hydrochloride Loperamide hydrochloride Fentanyl citrate Methadone hydrochloride Propoxyphene hydrochloride Pentazocine Levorphanol tartrate.
  • Narcotic antagonists: Nalorphine hydrochloride, Levallorphan tartrate, Naloxone hydrochloride.
